Why does the vortexer continue to run for approx. 2 seconds after relieving pressure on the vortex mat?
This 2-second overrun makes the vortexing process more ergonomic, because the vortex mat need not be pressed continuously to keep the vortexer running. It also eases the process of performing vortexing in several tubes successively.

How can the temperature accuracy be verified?
How can the temperature accuracy be verified?
The best possible temperature accuracy can be guaranteed by individually adjusting a SmartBlock. Optimized temperature control for each block type keeps temperature overshoots and deviations caused by external interferences to a minimum. To guarantee optimal performance over the entire period of use, Eppendorf offers a preventive maintenance program and certification services: your Eppendorf ThermoMixers, ThermoStat C as well as the SmartBlocks are regularly cleaned, checked and adjusted. Additional information is available at: https://www.eppendorf.com/epservices
. Customers can also verify the temperature accuracy by themself. Eppendorf provides for that purpose the Temperature Verification System – Single Well for Eppendorf Mastercyclers (0056 000.003) in combination with the 384 well Sensor (0056 002.006).

Can PCR plates be used in SmartBlock plates respectively ThermoMixer FP?
SmartBlock plates and ThermoMixer FP (with the fixed SmartBlock plates) are dedicated for using MTP’s, DWP’s and also other skirted plates which meet the SBS standard. For optimal usage of PCR plates the SmartBlock PCR 96 and PCR 384 in combination with ThermoMixer C or ThermoStat C is the proper choice. These SmartBlocks possess conical bores which surround the conical wells of PCR plates properly for a perfect temperature transition.
What does the term 2DMix-Control technology describe/stand for?
What does the term 2DMix-Control technology describe/stand for?
2DMix-Control technology describes the ability of the MixMate, ThermoMixer C and ThermoMixers F to mix small sample volumes in various plate and tube formats quickly and in a controlled manner. To achieve this, the interplay of all crucial factors for efficient and controlled mixing, have been optimized. And these are:
- Generation of sufficient mixing energy: due to an orbital mixing movement at a high mixing speed and a relatively large mixing radius (1.5 mm) the devices generate enough mixing energy (analogous to g-force in centrifuges) to mix even the smallest volumes efficiently.
- Minimized loss of mixing energy: special attention was paid to reducing inherent vibration. Smooth operation combined with reliable stability ensures that all the mixing energy is available for mixing and not wasted through transmission of vibrations to the device or its surroundings.
- Controlled transmission of energy from the device to the sample: Minimizing vertical movements enable a plane, 2-dimensional (2D) mixing stroke. This, in conjunction with the reliable plate and tube holders and SmartBlocks, prevents uncontrolled chaotic movements and allows all formats to be held securely, even at high mixing frequencies.

What types of applications can be carried out with Eppendorf Tubes® 5.0 mL?
What types of applications can be carried out with Eppendorf Tubes® 5.0 mL?
Basically, this tube type can be used for the same applications as the micro centrifugation tubes available in 1.5 and 2 mL formats. They close a gap between this small tube format and conical 15 mL tubes for sample volumes up to 5 mL. The tubes can be used in the field of cell culture. For this purpose, they are available in the purity grades “Sterile” (= sterile and pyrogen-free) or Biopur®. They are suitable for lysis of cells and tissues and purification of nucleic acids and proteins. For this, the PCR clean purity grade and LoBind variants are provided. They are also ideally suited for the preparation of large volumes of master mixes, buffers or dilution series as well as for space-saving storage of samples.

Up to which g-force can the Eppendorf Tubes® 5.0 mL be centrifuged?
All tube types are resistant up to 25,000 x g when used in combination with a suitable rotor or adapter. (Test conditions: 45° fixed-angle rotor at 40 °C, with aqueous salt solution (density 1.0 g/mL) and 90 mins). The mechanical loading capacity of the tubes is reduced by the use of organic solvents.
For which applications may I use Eppendorf Conical Tubes® 25 mL?
For which applications may I use Eppendorf Conical Tubes® 25 mL?
In general, all applications where standard conical tubes 15mL and 50 mL are routinely used. The Eppendorf Conical Tubes® 25 mL close a gap between 15 mL and 50 mL conical tubes and additionally offer in this format a unique snap cap – the so called SnapTec™ cap - which markedly improves handling. This makes the Eppendorf Conical Tubes® 25 mL ideally suited for all cell/tissue culture applications including splitting cells, media change, transfections, etc. Also assay preparation and biomolecule purification protocols (gDNA, plasmid DNA, RNA, proteins), which require organic solvent extractions and multiple centrifugation steps may be successfully performed. They are also ideally suited for general reagent / sample handling and space-saving storage.
Is it necessary to buy new equipment to enable the use of Eppendorf Conical Tubes 25 mL…
Is it necessary to buy new equipment to enable the use of Eppendorf Conical Tubes 25 mL within existing lab periphery?
No. The bottom shape / diameter of the Eppendorf Conical Tubes 25 mL is virtually the same with standard conical 50 mL tubes. With the respective height adapter (provided as Eppendorf accessory), the Eppendorf Conical Tubes 25 mL will be compatible with a wide range of standard laboratory equipment: centrifuges, mixers, etc., additionally tube racks, storage boxes and single-tube stands (skirts) are available for lab bench work, weighing and storage. Correct opening and closing of the snap Lid (SnapTec®).
Correct opening and closing of the snap Lid (SnapTec®).
The snap cap of the Eppendorf Conical Tubes® 25 mL is much larger than in other tube formats and may require some short learning of correct closing/opening at the beginning. Opening: press the safety tab from below and steady press upwards. In case of pressure buildup (organic solvents, higher temp incubations) make sure you slightly hold the lid with your thumb and don’t let the lid to pop out rapidly. Closing: press down the lid in the front area and make sure the safety tab clicks and is locked at right angle to the tube. Improperly locked tab may cause leaking.
Smart Block cryo Thaw on ThermoMixer C - What is the ‘Thawing cells’ program?
Smart Block cryo Thaw on ThermoMixer C - What is the ‘Thawing cells’ program?
The 'Thawing cells' program is an optimized program to thaw mammalian cells in 1.8-2.0 mL cryovials:

reproducibly
contamination free
standardized

It is a ready-to use program, in most cases no changes are required. The pre-set time is 3 minutes (can be extended up to 5 minutes), the pre-set mixing frequency is 500 rpm (can be adapted between 300 – 800 rpm), the temperature profile is fixed.

The settings are based on tests with standard mammalian cell lines (CHO, HEK, S16) up to highly sensitive stem cells (hiPSCs) at tested conditions with 2.0 mL cryovials / 1.0 mL fill (1 – 5 vials in parallel).

Other volumes or more vials may require an adaption of thawing time.

Further information can be found in the Application Note 437.
What is the difference between the SmartBlock cryo thaw and the SmartBlock cryo?
What is the difference between the SmartBlock cryo thaw and the SmartBlock cryo?
The 'SmartBlock cryo thaw' replaces the 'SmartBlock cryo'. Newly implemented is an automatic function for thawing cells (program 'Thawing cells') which is available in combination with the ThermoMixer C.
