Is the housing of ThermoMixer C, ThermoMixer F and ThermoStat C stable against UV irradiation?
A disinfection by UV irradiation cannot be recommended, because discoloration of the housing materials can occur.
Which DNA and RNAse deconatmination agents can be recommended for decontamination of the devices and the SmartBlocks?
We tested DNA away and RNase away and we can recommend to use both agents.
Are start and end position of the SmartBlocks the same?
No. Start and end position are not identical.
How can the temperature accuracy be verified?
The best possible temperature accuracy can be guaranteed by individually adjusting a SmartBlock. Optimized temperature control for each block type keeps temperature overshoots and deviations caused by external interferences to a minimum. To guarantee optimal performance over the entire period of use, Eppendorf offers a preventive maintenance program and certification services: your Eppendorf ThermoMixers, ThermoStat C as well as the SmartBlocks are regularly cleaned, checked and adjusted. . Customers can also verify the temperature accuracy by themself. Eppendorf provides for that purpose the Temperature Verification System – Single Well for Eppendorf Mastercyclers (0056 000.003) in combination with the 384 well Sensor (0056 002.006).
Can cell culture plates be used in the SmartBlock plates?
Also cell cultuture plates can be used in the SmartBlock plates in so far the dimensions of the plate comply to the SBS standard.
Is it possible to put the SmartBlocks for a rapid cool down into a -20°C freezer?
The SmartBlocks can be used at ambient temperatures of 5°C – 40°C and can be cooled appropriate. Cooling down of SmartBlocks in a freezer should not be done.
Is it possible to run more than one program in automatic succession on a ThermoMixer or ThermoStat C?
No. Each program has to be chosen and run separately. But the 4 possible steps within a program run in automatic succession.
Which SmartBlocks and vessels can be used with ThermoMixers F?
The ThermoMixers F possess a fix installed SmartBlock and can only be used with the appropriate microtest tubes or plates, such as MTP’s, DWP’s or skirted plates complying the SBS standard. For using PCR plates our SmartBlocks PCR 96 and PCR 384 in combination with ThermoMixer C is the proper choice.
Do the IsoRacks fit upon the SmartBlocks for microreaction tubes (0.5 mL; 1.5 mL; 2.0 mL)?
No. The IsoRacks fit only upon the Thermoblocks for the older devices Thermomixer comfort, Thermomixer compact & ThermoStat plus.
Can PCR plates be used in SmartBlock plates respectively ThermoMixer FP?
SmartBlock plates and ThermoMixer FP (with the fixed SmartBlock plates) are dedicated for using MTP’s, DWP’s and also other skirted plates which meet the SBS standard. For optimal usage of PCR plates the SmartBlock PCR 96 and PCR 384 in combination with ThermoMixer C or ThermoStat C is the proper choice. These SmartBlocks possess conical bores which surround the conical wells of PCR plates properly for a perfect temperature transition.
What does the term 2DMix-Control technology describe/stand for?
2DMix-Control technology describes the ability of the MixMate, ThermoMixer C and ThermoMixers F to mix small sample volumes in various plate and tube formats quickly and in a controlled manner. To achieve this, the interplay of all crucial factors for efficient and controlled mixing, have been optimized. And these are:
- Generation of sufficient mixing energy: due to an orbital mixing movement at a high mixing speed and a relatively large mixing radius (1.5 mm) the devices generate enough mixing energy (analogous to g-force in centrifuges) to mix even the smallest volumes efficiently.
- Minimized loss of mixing energy: special attention was paid to reducing inherent vibration. Smooth operation combined with reliable stability ensures that all the mixing energy is available for mixing and not wasted through transmission of vibrations to the device or its surroundings.
- Controlled transmission of energy from the device to the sample: Minimizing vertical movements enable a plane, 2-dimensional (2D) mixing stroke. This, in conjunction with the reliable plate and tube holders and SmartBlocks, prevents uncontrolled chaotic movements and allows all formats to be held securely, even at high mixing frequencies.
Is it possible to use the Eppendorf Thermomixer for 24 h and at 7 days?
Yes, this is possible. The customer has just to set the time to endless.
What types of applications can be carried out with Eppendorf Tubes® 5.0 mL?
Basically, this tube type can be used for the same applications as the micro centrifugation tubes available in 1.5 and 2 mL formats. They close a gap between this small tube format and conical 15 mL tubes for sample volumes up to 5 mL. The tubes can be used in the field of cell culture. For this purpose, they are available in the purity grades “Sterile” (= sterile and pyrogen-free) or Biopur®. They are suitable for lysis of cells and tissues and purification of nucleic acids and proteins. For this, the PCR clean purity grade and LoBind variants are provided. They are also ideally suited for the preparation of large volumes of master mixes, buffers or dilution series as well as for space-saving storage of samples.
Is it necessary to buy new equipment to be able to use the new tube?
The bottom shape of the Eppendorf Tubes® 5.0 mL is similar to that of conical 15 mL tubes which means that they are compatible with a wide range of standard laboratory equipment. In addition, an extensive range of accessories is available, enabling them to be used in many devices. Both racks and storage boxes are available for lab bench work and storage. Incubation and mixing operations can be performed using suitable blocks on Eppendorf thermomixers and compatibility with Eppendorf centrifuges is ensured by special rotors and a variety of adapters. With a suitable reservoir rack module, the tubes can also be integrated into the epMotion® automated liquid handling workstation. Is it possible to store samples in 5.0 mL Eppendorf Tubes® in the freezer?
Yes, the tubes can be stored down to -86 °C.
How resistant are the Eppendorf Tubes® 5.0 mL to chemicals?
The tubes are made of high quality highly transparent polypropylene and therefore resistant to a variety of commonly used chemical substances in the laboratory. An overview is provided in Application Note No. 56.
Are the Eppendorf Tubes® 5.0 mL available with the same purities and LoBind variants as the small micro centrifugation tubes from Eppendorf?
The Eppendorf Tubes® 5.0 mL are available in the purity grades Eppendorf Quality, PCR clean and Biopur® and also in the purity grade Sterile. In addition to irradiation for sterilization, this also includes certified testing for the absence of pyrogens. The tubes are also available as DNA LoBind and Protein LoBind variants.
Can the universal adapter for conical 15 mL screw-cap tubes be used in each rotor/adapter that is designed for this tube type?
It can be used in the respective Eppendorf rotors/adapters, but we assume no liability for use in rotors/adapters from other manufacturers.
What is the Tube Clip used for?
The Eppendorf Tubes® 5.0 mL offer secure closure at incubation temperatures up to 80 °C. For higher temperatures, the Tube Clip should be used to secure the lid.
Up to which g-force can the Eppendorf Tubes® 5.0 mL be centrifuged?
All tube types are resistant up to 25,000 x g when used in combination with a suitable rotor or adapter. (Test conditions: 45° fixed-angle rotor at 40 °C, with aqueous salt solution (density 1.0 g/mL) and 90 mins). The mechanical loading capacity of the tubes is reduced by the use of organic solvents.
How is the FN1 motifs surface made?
Based on a proprietary coating technology, the FN1 motifs surface is made up of synthetic fibronectin-derived motifs (including RGD peptide sequence), specifically designed to mimic the cell attachment site of native extracellular matrix proteins.
Is FN1 motifs a two-dimensional surface?
Yes, FN1 motifs is a two-dimensional surface.
What peptide sequences are used to mimic the ECM?
The FN1 motifs surface is made up of synthetic fibronectin-derived motifs including RGD peptide sequence.
Is FN1 motifs also available for manual coating?
A FN1 motifs substrate for manual coating is currently not available.
How is FN1 motifs manufactured?
The FN1 motifs consumables are developed and manufactured according to ISO 9001. Production of all Eppendorf Cell Culture Consumables takes place in a controlled class 5 clean room environment according to ISO 14644-1 and EG-GMP Class A. They are manufactured using aseptic processing per ISO 13408-1 “Aseptic processing of healthcare products”. Each step was validated to ensure the FN1 motifs consumables meet the Sterility Assurance Level (SAL) 10-3. The consistent reliable quality of FN1 motifs consumables is ensured by monitored process controls and checks from the raw material to the final packed product. The growth surface is ensured to fulfill all requirements concerning homogeneous coating and to be animal- and xeno-free.
Does FN1 motifs have autofluorescence?
No, FN1 motifs cultureware does not present autofluorescence.
Do I have to prepare the FN1 motifs surface before use?
No, the FN1 motifs surface is ready-to-use and does not require any pre-equilibration or washing step before use. Keep the product sealed in original packaging and open it just before use.
I have already opened the package. How soon do I have to start working?
The package should be opened just prior to use.
Why is the FN1 motifs surface individually packed in aluminum foil?
The aluminum foil protects the FN1 motifs surface from the external environment (e.g. UV light) and ensures its stability and sterility throughout the shelf-life of 3 years at room temperature based on unopened product.
The FN1 motifs consumable is exposed to the laboratory environment without the aluminum-foil based packaging. Can I still use the surface?
We do not recommend using the product after vessels are exposed to the laboratory environment without the aluminum-foil based packaging. Stability and sterility cannot be guaranteed anymore.
