Increasing Upstream Bioprocessing Efficiency through Process Analytical Technology

In biologics production, the upstream bioprocess contributes significantly to product quality and manufacturing efficiency. A comprehensive understanding on which process parameters are critical and how they influence process outcome allows the development of tailored control strategies and, as a result, achieving consistent process performance.

Compared to offline measurements in a sample that has been taken from the bioreactor, inline analysis of process parameters reduces the manual workload and facilitates the implementation of automated feedback control loops. While inline analytics of some process parameters, such as temperature, DO, and pH is well established, real-time measurement of others, such as metabolite concentrations or cell viability is not yet routine.

Here we present strategies for in line process analytics that aimed at automated feed control in an upstream bioprocess. Using the example of a Raman analyzer, a glucose analyzer, and an exhaust analyzer we demonstrate, how to achieve seamless sensor integration with the bioprocess control software. In all cases we met the challenge of keeping the glucose concentration stable during the bioprocess run and minimize human intervention.

The Potential of Hypoxic Culture in Large Scale Expansion of MSC for Therapeutic Purpose in Regenerative Medicine

MSC production is an expensive and high cost process with a high price to pay for any failure in MSC production. Conventional culture of Mesenchymal stem cells (MSC) either in the lab or cGMP facility is mostly in an in the environment with 21% oxygen level. However, stem cells in human tissues live in an environment with much lower oxygen concentration (<5%).

Muchos de esos fármacos son anticuerpos monoclonales humanos o humanizados. El objetivo del presente webinar es mostrar los resultados de tres principales métodos de producción para un biosimilar típico (lote, lote alimentado y perfusión) y su ejecución fácil y sencilla en un sistema de bioprocesos de mesa, lo que permite tomar decisiones confiables sobre el proceso para que sea escalable a producción.

Cell Culture Bioprocess Scale-Up Workflow from Bench to Pilot/Production Scale using Single-Use Bioreactors

Transferring a biologic candidate from the research and development phase to commercial production usually requires increasing the working volume of the upstream bioprocess. During scale-up, process performance optimized at small scale needs to be reproduced at larger scales, ideally without much need for process optimization at large working volumes. This requires reproducing the quality of the seed culture and the cells’ growth environment across scales.

To streamline scale-up, bioengineers commonly use bioreactors with similar geometries at all scales and keep one or more parameters constant between vessels of different sizes, such as kLa, power input per volume or tip speed. Therefore, to design a scale-up strategy, data on scale-up-relevant engineering parameters need to be available for each bioreactor planned to be used. These parameters include vessel geometries, power numbers, and oxygen transfer capabilities.

In this webinar we showcase scale-up of an antibody production process in CHO cells from bench to pilot/production scale. We discuss strategies to prepare seed cultures of reproducible quality in shake flasks. We demonstrate a new concept for software-aided scale-up strategy design, which considers vessel parameters of differently sized Eppendorf BioBLU® Single-Use Vessels and Thermo™ HyPerforma™ Single-Use Bioreactors.

Tailoring oxygen transfer: A key driver for bioprocess intensification

An appropriate dissolved oxygen (DO) concentration in the culture medium is a key driver for high bioprocess yields. DO is determined by the oxygen supply to the bioprocess system and the oxygen consumption of the cultured microorganism simultaneously. In high-density fermentation of aerobic bacteria and fungi, for example, a large amount of oxygen must be introduced into the medium to avoid oxygen limitation during robust microbial growth. In other applications, like the cultivation of microaerophilic bacteria for probiotics production or gut microbiome research, the DO concentration must be controlled precisely at a low level.

In this webinar, we showcase advanced strategies to control the dissolved oxygen concentration in the culture medium and to reproduce it at different scales.

Optimize your bioprocess – From shaker to bioreactor

Bioreactors and shakers are used to cultivate microorganisms, plant, insect, and mammalian cells in different volumes. Upscaling of production often involves shake flasks and bioreactors. Shake flasks are used for bioreactor inoculum production and are an easy-to-use instrument for early process development like organism screening and media design. At the next process steps, highly controllable bioreactors are needed for flexible solutions, automated process control, and predictable scalability when larger quantities of cells are required. The challenge during upscaling is to keep a constant efficiency and quality during all process steps.

Stem Cell Derived Exosome Production in Stirred-Tank Bioreactors

Exosomes are a population of naturally occurring mobile, membrane-limited, 30 – 100 nm in diameter, extracellular vesicles containing a large number of proteins, lipids, messenger, and micro-RNAs. It was shown that they play a role in the mediation of intercellular communication, the modulation of immune-regulatory processes, tumor metabolism, and regenerative as well as degenerative processes. In recent years, there is increasing interest in the therapeutic potential of exosomes produced by Mesenchymal Stem Cells (MSCs).

Stirred-tank bioreactors offer the possibility to tightly control and monitor the production of exosomes, as well as the scalability, to produce increasing amounts. However, the cultivation of stem cells in stirred-tank bioreactors requires profound knowledge and precise control of the process.

In our talk, we will highlight the potential and benefits of stirred-tank bioreactors in the cultivation of stem cells for exosome production. We will share our knowledge on how we developed the process in our DASbox® Mini Parallel Bioreactor System and scaled-up the process to 1 L controlled by our newest parallel bioreactor control system, the SciVario® twin.

Anchorage-dependent cells, such as Vero cells, are widely used as a platform for viral vector production. To cultivate Vero cells in stirred-tank bioreactors, they require a growth matrix to attach to. From small-scale stem cell production to large-scale hiPSC-based drug development: The flexibility of stirred-tank bioreactors

Stirred-tank bioreactors are gaining more and more importance in the industrial production of pharmaceutical products, biofuels and food. When compared with 2D-cultivation systems, bioreactors are easy to handle, and multiple bioreactors can be controlled in parallel without a considerable increase of workload. In stirred-tank bioreactors cells can be cultivated as free cells in solution, cell aggregates or on microcarriers in batch, fed-batch or continuous cultivation modes.

The first part of this webcast will feature a review of the basic principles of stirred-tank bioreactors. Ankita Desai of Eppendorf North America will explain factors that must be considered before and during the process, how to set up perfect conditions for cells, and how the shape of the impeller influences the stem-cell culture.

Within the second part, Arie Reijerkerk and Farbod Famili from Ncardia, a stem cell drug discovery and development company, will provide insight into how the advent of human-induced pluripotent stem cell (hiPSC) technology has substantially expanded the availability of human cells for drug discovery. The availability of these cells helps researchers to uncover the potential for better translation to the clinic and reduction of late-stage drug attrition.

Ncardia will present a case study of large-scale manufacturing development and their hiPSC-based drug screening platform, DiscoverHIT, including results from the generation of disease-relevant human stem cell-derived models and the development of predictive, phenotypic assays to enable high-throughput screening.

The emergence of NGS is revolutionizing the microbiological sciences and transforming medicine. Deep sequencing has revealed virtually all environments, including the human body, are teeming with diverse microbial communities. Culture of 3D Cell Aggregates in Perfusion

Three-dimensional (3D) cell aggregates are of great interest for many applications, including disease modeling, drug toxicity assessment, and manufacture of stem cell-based products. Stirred-tank bioreactors are promising culture systems for 3D cell aggregates, as they allow efficient establishment and maintenance of cell aggregates, process monitoring and control, and process scale-up to larger volumes. Furthermore, they can be operated in perfusion mode, which allows 3D cell aggregates to be sustained longer than in traditional batch cultures.

This webcast will review a research example for process development with the human tumor cell line H157, cultivated in stirred-tank mini bioreactors as 3D cell aggregates.

Expansion of Human Stem Cells in BioBLU® Single-Use Vessels from Eppendorf

Experimentation involving human stem cells (HSCs) is one of the fastest growing fields in research and development due, in large part, to its potential for revolutionizing human disease treatment.  There are multiple HSC platforms being investigated, including human mesenchymal stem cells (hMSCs) and human induced pluripotent stem cells (hiPSCs). Both have distinct advantages and disadvantages, but what they share is a bottleneck at the stage of scale-up, or the ability to produce enough cells for downstream R&D purposes. In this study, we discuss the successful growth of HSCs, both hMSC and hiPSC, in BioBLU® single-use stirred-tank bioreactors from the small (0.3 L) to bench (3 L) scale. In particular, we highlight the production of one billion hMSCs in a single bioreactor, and the culture of hiPSC at the small scale in parallel bioreactors.

The Tip of the Iceberg – How Pipette Tips Influence Asssay Results

We looked below the waterline and got unexpected findings

When was the last time you were upset about non-reproducible results of your experiment? Even though you checked all variables including the pipetting system used in your experiment, you were unable to find the cause.

In many cases the assumption is made that the pipetting system, consisting of pipette and pipette tips, cannot be a source of error as long as the tip fits the pipette. However, even if this is the case, this does not speak to the precision and accuracy of the pipetting result. The international standard ISO 8655:2002 considers pipettes and pipette tips as a system. With good reason!

In this webinar we will show the results of our comprehensive study about tip quality in system with the pipette. At the end of this webinar you will have an overview of several influencing factors impairing your pipetting results when using pipettes and tips. We found that in the same way that only the tip of an iceberg can be seen above water level, a number of other problems with pipette tips rather stay unknown.

Optimizing a Biosimilar Production Process

The global market for biosimilars is expected to reach US$25 bn by 2020, when more than ten global blockbuster biopharmaceuticals, with sales of more than US$67 bn, will be off-patent. Many of these drugs are human or humanized monoclonal antibodies (hMAbs).

The objective of the research project presented in this webinar was to determine if the three primary methods of production for a typical biosimilar (batch, fed-batch, and perfusion) may be simply and easily executed using a single, benchtop bioprocess control system to allow confident process decisions that are scalable to full production. We compared two perfusion processes. For the fed-batch and perfusion cultivations, we used stirred-tank single-use bioreactors.

This webinar will discuss the outcome of these studies, and will provide an analysis of the benefits of their respective usage in biosimilar production with a particular focus on improving time and cost efficiency.

Good Plastics, Bad Plastics – How Tubes, Plates and Pipette Tips Can Affect Your Experiments

Substances leaching out of plastic consumables – so-called “leachables” – are still frequently underestimated in laboratory workflows. While possible effects of leachables are routinely taken into account in pharmaceutical or food research, this topic is still underrepresented in the majority of life science applications. Recent scientific evidence shows however that leachables can significantly affect experiments and pose a likely source of error in many assay systems for example, by inhibiting enzyme activities in bioassays or falsifying nucleic acid quantification.

Controlling Expansion and Cardiomyogenic Differentiation

To harness the potential of human pluripotent stem cells (hPSCs), abundant provision of their differentiated progenies is required. During this webinar we present how hPSC expansion as matrix-independent aggregates in suspension culture (3D) was successfully combined with cardiomyogenic differentiation. A multi-well screening approach was scaled-up to stirred-tank bioreactors applying controlled feeding strategies (Batch and Cyclic Perfusion) followed by differentiation.

Optimized conditions in 100 mL cultivations enabled the production of 40 million cardiomyocytes (>80% purity), the majority of which displayed ventricular-like action potentials and were directly applicable for bioartificial cardiac tissue formation, a potential strategy for heart repair, drug discovery and safety pharmacology.
