The Concentrator plus has various functions for the concentration of samples or for the removal of solvents. How is it used?
You can operate the Concentrator plus as a concentrator, desiccator or as a centrifuge. There are three different modes for various solvents available for the concentrator or desiccator function respectively: for aqueous, for alcoholic solvents and for solvents with high vapor pressure.
The concentrator function enables the quick concentration of samples via vacuum (fast evaporation of the solvent) and centrifugation (prevents foaming over/sample loss) in the rotor. The desiccator function enables fast drying under vacuum conditions without using a rotor, e.g. for filter papers. The sample can be placed directly in the rotor chamber; the rotor shaft doesn't rotate during the run. With the centrifuge function, the rotor spins, but the rotor chamber is not evacuated.
How can I set the run time in the Concentrator plus?
The run time can be set with the time arrow keys. The setting of 1 min to 9:59 hours and endless (∞) is possible.
What temperatures can one set with the Concentrator plus?
The temperature steps --, 30, 45, 60 °C can be set using the temp key and are visible on the display, whereby -- symbolizes evacuation st room temperature. The temperature can be changed during operation. The new temperature is immediately readjusted.
What braking functions does the Concentrator plus have?
The braking function can be set with the brake key. ON is found above this function key on the display when the brake is switched on. The rotor is in this way gently decelerated at the end of the run. Deceleration can also take place without braking. For this purpose push the brake key until OFF appears in the display. It is possible to move between these settings at any time.
Is it possible to change the set mode in the Concentrator plus during the run?
The braking function, the temperature and the total run time can be changed. The new parameters are adjusted immediately. During the run, the set function, which can be selected with the mode/vent key, cannot be changed.
Is it possible to ventilate the Concentrator plus during the run?
The ventilation valve is opened by pressing the mode/vent key during the run. Air flows in and rinses the pump and tube system. Prior to concluding daily work and upon finishing the last run, we recommend pressing the button once for a few seconds in order to remove residual condensation from the pump.
Is there a recommended ambient temperature for the operation of the Concentrator plus?
The ambient temperature for the operation of the Concentrator plus complete system may range from 15 °C to a maximum of 35 °C.
What rotational speeds can be set in the Concentrator plus and which g-force does this correspond to?
The rotational speed for all rotors is 1,400 rpm. It cannot be changed. The g-force varies according to the position of the tubes in the rotors between 130 and 250 x g.
Can I retrofit the Concentrator plus basic device into a complete system?
No, an upgrade of the Concentrator plus basic device is not possible.
Which properties must a vacuum pump have in order to be able to connect it to the Concentrator plus basic device?
In order to ensure the functionality of the Concentrator plus, an external vacuum pump must have a suction capacity of at least 1.9 m³/h at 50 Hz or 2.1 m³/h at 60 Hz. Each pump used should be able to contain a maximum ultimate pressure of at least <20 hPa (<20 mbar). The vacuum pump should also be resistant to chemicals. Additional information are listed below the tab "notes & papers" at our user guide 030.
How does one connect a vacuum pump to the Concentrator plus basic device?
An external vacuum pump with a power input of < 400 W can be connected directly to the basic device with a special plug. A socket is found at the rear of the device. In contrast, an external vacuum pump with a power input of > 400 W must be connected directly to the power supply. The control of the pump by the Concentrator in this case takes place via a solenoid valve located between the vacuum pump and the Concentrator plus. What material is the membrane vacuum pump of the Concentrator plus made of?
The inside of the pump is coated with polytetrafluorethylene (PTFE). The feeder from the rotor chamber to the pump is, in the case of the Concentrator complete system, also coated with PTFE .
What material is the lid of the Concentrator plus made of?
The lid is of polymethylmethacrylate (PMMA). All devices sold from September 2013 on are equipped with a new coated lid, which provides superior chemical resistance.
What cleaning method do you recommend for the Concentrator plus?
Only neutral, gentle cleaning agents and alcohol-based disinfectants may be used for the cleaning and disinfection of the device. This applies for both the housing and the rotor chamber. The mains plug must be disconnected prior to cleaning. It should principally be prevented that aqueous, saline solutions be allowed to dry, and the longer term effects of acids or lyes on the material avoided.
What cleaning method do you recommend for the rotors of the Concentrator plus?
Neutral, gentle cleaning agents and alcohol-based disinfectants may be used for cleaning and disinfection. Where contamination with biologically dangerous elements is suspected, we recommend autoclaving the rotors (121 °C, 20 min).
What purpose do the feet of the rotors F-45-48-11 and F-45-72-8 serve of the Concentrator plus?
The small metal feet can be screwed into the corresponding bores of these rotors. They ensure that the tubes are not pushed out of the bores when one places the rotor on the workbench. The rotors can be used in the Concentrator plus with the feet. They should be regularly tightened.
Why should a warm-up phase be carried out prior to starting with actual work with the Concentrator plus?
The membrane vacuum pump only reaches the suction capacity and ultimate pressures given in the "Technical Data" after a warm up phase of the Concentrator of approx. 15 min. The warm-up phase reduces the condensation of liquids in the pump and on the lid of the rotor chamber. If condensation should nonetheless accumulate in the pump after large sample throughput, please let the Concentrator run for several minutes following the completion of work.
How quickly can samples be evaporated in the Concentrator plus?
The time required for the concentration of a sample depends on the properties of the sample (vapor pressure, boiling point, etc.), the quantity of liquid to be concentrated and on the type of vessel.
What chemicals can be concentrated with the Concentrator plus?
The Concentrator was developed to concentrate or dry samples with DNA, RNA, nucleotides and proteins, which are found in alcoholic or aqueous solutions.
Is it possible to evaporate ether with the Concentrator plus?
No. The Eppendorf Concentrator plus may not be used in a potentially explosive environment and must be operated in a safe, adequately ventilated environment or beneath an extraction hood. Chemicals that can potentially create an explosive atmosphere may not be used in the Concentrator plus.
Is it possible to connect a cold trap to the Concentrator plus?
You can connect any commercially available cold trap to our Concentrator plus. The cold trap can be used behind the pump in place of the emission condenser with the supplied tube.
In which cases is it absolutely necessary to use a cold trap?
When concentrating flammable, chemically aggressive liquids or those with volatile, chemically aggressive or biologically hazardous components, an effective cold trap or chemical trap must be used in place of the emission condenser. Only this way users can be protected from solvents that are dangerous to health. In the case of the Concentrator complete system, the solvent-resistant pump allows the setup behind the pump.
When using a cold trap, must a special tube be used for connecting the pump with the cold trap?
The PVC tube supplied by Eppendorf, which normally connects the pump and the emission condenser, can be used, for chemicals to which PVC is resistant.
Can you recommend a cold trap manufacturer?
According to our information, the following cold trap can be recommended: COLD TRAP type KF 29-GL incl. Dewar from the KGW-Isotherm company. An overview of dealers and representatives can be found at their homepage
Can 0.2 ml tubes be used in the Concentrator plus?
Yes, one can centrifuge 0.2 mL tubes in the 1.5 mL rotor F-45-48-11 with the help of an adapter #5425 715.005 (order no North America #022636260), in the 0.5 mL rotor F-45-72-8 with the help of the adapter #5425 723.008 (order no North America #022636286).
How high may Microtest Plates be in order to use them in the swing-bucket rotor A-2-VC?
The height of 27 mm for Microtest Plates may not be exceeded.
To what height may the Microtest Plates be filled?
During centrifugation, the menisci stand angled in the tubes at the edge of the plates. This is caused by the centrifugal force and is unavoidable. Therefore, the wells of the plates should only be filled to a maximum of approx. 2/3 of the max. filling volume.
How can I work with two rotors at the same time (sandwich operation) in the Concentrator plus?
When a spacer (#5301 316.005, (order no North America #022822101) is placed between them, two rotors can be centrifuged simultaneously (possible with the rotors F-45-48-11 and F-45-72-8 in desired order, not with rotor F-45-70-11).When using in combination, the maximum load is reduced:
• Lower rotor: 2/3 of nominal maximum.
• Upper rotor: 1/3 of nominal maximum.
Can filter plates be dried with a vacuum in the Concentrator plus?
Yes, the Concentrator plus can also be operated purely as a desiccator. The rotor chamber is thereby evacuated normally, but the rotor remains immobile. With the mode/vent key, depending upon the solvent with which the filter plate is treated, the functions D-AQ, D-AL or D-HV can be selected in the display.
What do I need to consider when working with the Concentrator plus complete system with junction for external devices (#5305 000.614) and a connected gel dryer?
On a complete system with junction for external devices, a gel dryer can be run in parallel to the operation as a concentrator/desiccator and/or used separately. For parallel work we recommend the V-HV and D-HV modes. If the gel dryer is run separately, you should use D-HV mode in order to protect the drive. In contrast to the concentrator or desiccator function, open the tap of the gel dryer connection before operation. The knob thereby points in the flow direction. Close it again after operation. When the gel dryer is operated without the concentrator or desiccator function the device lid must be closed as a vacuum otherwise cannot be generated. Then proceed as with the concentrator function. Please note the solvent properties when carrying out work. When running several devices in parallel on a vacuum system and/or in combination with a gel dryer, an explosive gas mixture can develop in the vacuum system. Only carry out parallel operations with identical solvents or solvents that evaporate in a non-hazardous manner.
