How does the shaker agitation rate and orbit affect culture growth?

Generally, agitation speed and orbit affect both the aeration and the mixing of the culture. Greater aeration increases oxygen transfer rate (OTR), boosting cell growth. The size of the orbit affects OTR linearly and the speed has a square-law relationship. Therefore, if the size of the orbit doubles, say from 1/2 to 1," the amount of OTR should double〖RPM〗_1/〖RPM〗_2 = (r_2^2)/(r_1^2 ), then 〖RPM〗_2 = √(〖RPM〗_1^2 (r_1/r_2 ) ). If the speed doubles, the OTR should quadruple. Of course, these are general rules and caution should be taken with shear sensitive cells. However, this information can be used to estimate the conditions needed to obtain the same results on different model shakers. For example, to get the same oxygen transfer in a shaker with a 1 in orbit running at 300 rpm, a shaker with 3/4 in orbit must be run at 350 rpm.

Do Eppendorf shakers come with platforms?

Each Eppendorf shaker is available with multiple platform options, which are ordered separately to give you the flexibility to choose the one that best suits your needs. Dedicated platforms include installed clamps of a single size (ie. only 250 mL clamps) and provide in most cases the best capacity but does not allow for the use of different size clamps or holders. Universal Platforms are designed for use with a combination of various-sized clamps and test tube racks providing maximum flexibility. When ordering a Universal Platform, clamps and test tube racks must also be ordered separately.

What is the temperature offset function?

The temperature offset function on many of our shakers allow you to match a reference point" temperature inside the chamber (culture medium) with the "Actual Value" displayed. The temperature offset function on Innova Incubated shakers allows you to monitor the temperature of the media providing you a clearer and more accurate picture. Please be aware that matching reference point temperatures to the Actual means you have to deal with many variables such as: greater evaporation; heat gains generated by cultures growing - (slowly at first and much more during their growth phase) and heat generated by the friction of media with the glass at high speeds or use of baffled flasks. Over time, the reference point temperature offset may need to change due to these varying conditions.

I want to grow my cultures at 27°C, do I need an incubated shaker with refrigeration?

The Eppendorf Incubated, non-refrigerated Shakers Innova and I-Series are designed to maintain temperatures of ambient plus 5 °C to its maximum operating temperature. The Eppendorf Incubated Excella shakers will operate from ambient plus 7 °C to maximum temperature. It is critical to understand that "ambient temperature is not the temperature on the wall thermometer (room temperature), but the temperature measured one meter from the front of the machine.

Why are some flask clamps supplied with an extra spring set?

This "extra spring set" (we call it a girdle) is included with clamps for 2 L, 2.8 L, 4 L and 6 L flasks. It is intended to be used with the already installed spring set. This girdle helps prevent large flasks from spinning inside the clamp and reduces the risk of flasks becoming dislodged or scored by the steel clamp. One girdle is already in place on the clamp, the other is packed separately. To install this extra “girdle”, follow the instructions in the manual supplied with the shaker.

How is temperature accuracy defined by Eppendorf?

Accuracy is defined as the degree of closeness between the displayed value and the actual value as determined by a traceable device. All Eppendorf Incubated Innova and I-Series shakers have a temperature Accuracy of ±0.1 °C from 30 – 40 °C, ±0.5 °C over the remaining range. New Brunswick Incubated Excella Shakers are accurate within 0.1°C at 37°C.

How is Uniformity defined by Eppendorf?

Uniformity is the ability to maintain uniform temepratures throughout the platform. New Brunswick Incubated Innova Shaker temperatures will not deviate by more than +/- 0.25 °C at 37 °C.

The orbit of a shaker is the distance traveled around the central drive. This can be descibed in inches, centimeters, or milimeters. If our shakers had a one inch (2.54 cm) orbit. That means that that the lateral movement of the platform would equal one inch from one point to another that is 180° opposite that position. All of the New Brunswick. shakers have a circular orbit

What wavelengths are used in your growth lamps in a photosynthetic light bank?

Each of the bulbs that is fit into our photobanks is a wide spectrum bulb that provides the red and blue wavelength of energy needed for photosynthesis. Innova 42R has six bulbs as the Innova 43R and 44R have nine bulbs. To have this option installed, the shaker needs to have the refrigeration option installed as well.

Which screws will fit to which platform ?

All of Eppendorf Inc. clamps and accessories are packaged with multiple screws to fix them to our universal platforms, included in M1013-0261 clamp screw kit (Stainless steel platform screw #S2117-3050 (10-32 x 5/16 in (7.9mm)), Stainless Steel, Aluminum, and Aluminum and Phenolic (Plastic) #S2116-3051 (10-24 x 5/16 in (7.9 mm)), and Wood #S2116-3101 (10-24 x 5/8 in (15.87mm)).

How can I disinfect, clean, maintain my shaker?

We recommend cleaning the exterior of our units with a damp cloth or any standard household or laboratory cleaner to wipe the surfaces clean. Do not use abrasive or corrosive compounds to clean the instruments as they may damage the unit and void the warranty. It is the responsibility of theuser to carry out the proper decontamination procedures if hazardous materials are spilled on or inside the equipment. Before using any cleaning or decontamination method other than those suggested by the manufacturer, users should contact Eppendorf Inc. to ensure that the proposed method will not damage the equipment. Commercially available household bleach solutions, when diluted at a 1:10 ratio, are effective in routine decontamination of the instrument. The method for decontaminating a spill depends on the nature of the spill. Spills involving fresh cultures or samples of known concentrations of biomass should be flooded with decontamination solution and soaked for at least 5 minutes before cleanup. Spills involving samples with high concentrations of biomass, or involving organic matter, or occuring in areeas warmer than ambient/room temperature, should be exposed to decontamination solution for at least one hour before being cleaned up. Personnel involved in the cleanup of any spill should wear gloves, safety glasses, and a laboratory coat or gown during the cleanup process. Respiratory protection should be considered for spills where aerosolization is suspected.

Why is the triple eccentric drive special ? What is the difference between Innova and Excella Shakers drive ?

The Innova and I-series Triple eccentric drive consists of three individual bearings and eccentric shafts mounted in a triangular fashion for precise uniform circular motion at every point on the platform. The heavy duty drive has a single shaft that utilizes springs to aid in balancing the load. This drive works great with small to medium loads at lower speeds.

How can I shake MTPs ? Which parameters ?

Eppendorf shakers offer a variety of Micro Titer Plate holders that attach to our univeral platforms. Our shakers offer a variety of orbits up to 5.1 cm (2 in) and speeds up to 500 rpm. If mixing is what is needed at higher speeds and smaller orbits you can find Eppendorf Mixers.

How do I use of sticky pads/sticky tape?

Eppendorf Sticky Pads are recommended for use on our sticky pad platform or our sticky pad adapter kit (M1250-9504). On the S44i universal platform sticky pad can be used directly. The adapter kit is easily fixed to our universal platforms. Any size flask can be used with the sticky pad with a 1 in (2.54 cm) orbit up to 250 rpm, any size flask can be used with the 2 in (5.1 cm) orbit up to 200 rpm. To clean the sticky pad, just wash with soap and water. this will revive" the stickiness, but the sticky pad must be completely dry before replacing on the platform.

What is the heavy load platform ?

The Heavy Duty Platform (M1282-9928) is designed for use in the Innova 44 only. This platform is 0.95 cm thick alluminum plate that is recommended for shaking full loads of flasks greater than 1 L at or above 20 % fill volumes or flasks with baffles.

Is there a minimal load required for the use of Eppendorf shakers?

The shaker must be used with a platform. This is the minimal load. Never use the shaker without an installed platform.

What is the orbit of a shaker?

An orbital shaker moves the platform in a circular shaking motion. The term orbit describes the diameter of this circular movement. Typical orbit sizes are 1.9 cm, 2.5 and 5 cm.

I use two shakers with different orbits. Can I transfer my experimental conditions to a shaker with a different orbit?

Yes, the rpm can be adapted with help of the following formula .

What platform types are offered with the shakers?

Universal platform and a variety of dedicated platforms are available. University platforms offer maximum flexibility having a hole-pattern that allows using virtually any combination of clamps and other accessories. Dedicated platforms come complete with pre-installed clamps of one size. The hole-pattern of the dedicated platform is optimized for the use of one specific size of clamps. It allows in most cased more capacity but can only be used with that size of clamp. Please note that the platform has to be ordered in addition to the shaker.

Is stacking with Innova 44 possible?

Yes, that is possible. Stacking requires a base and the specific stacking kit. For a double stack the S44i must be placed on top of Innova 44 independent of orbit. For a triple stack: If two S44i with different orbits are stacked on one Innova 44, place the one with larger orbit in the middle. If one S44i is refrigerated place that one in the middle. Note that devices only run speed reduced: 2 devices stacked: 20 rpm – 300 rpm. 3 devices stacked: 20 rpm – 250 rpm. Due to speed limitation Balance master Calibration should not be run. Initial counterweight settings can be taken from the manual.

What is the maximum flask size I can use in the S44i?

Erlenmeyer flasks up to 5 L in size can be accommodated. If the static Incubation shelf is installed, flasks up to 1 Liter Erlenmeyer or 2.8 / 3 L Fernbach can be accommodated. General capacity tables are available in the operating manual here .

Can I use all Eppendorf shaker accessories as well for the S44i?

Yes, all shaker accessories can be used with the S44i.

Can I use the S44i with sticky pad?

Yes, sticky pad can be used directly on the S44i Universal platform. The universal platform fits 9 sticky pads to be covered.

Can I run a double or triple stacked unit fully loaded and at full speed?

Yes, Balance Master function allows the optimal adjustment of the counterbalance to run all devices in a triple stack at maximum speed (300 or 400 depending on orbit) even with full load.

What is the maximum load on the platform?

The maximum load on the platform is 35.4 kg (78 lb).

What disinfection agents can I use to disinfect the S44i?

70 % Ethanol or Isopropanol or other suitable commercial disinfectant can be used for wipe disinfection the interior and exterior surfaces. For detailed information please refer to the operating manual at www.eppendorf.com .

Can I upgrade my shaker later on with other options when I buy a basic non-refrigerated unit?

Yes, the options photosynthetic LED lights (on refrigerated units only), the static shelf and the window shade can be upgraded to a unit. Refrigeration cannot be upgraded later on.

What can I do when I have spillages to the chamber bottom?

Depending on the amount of spill open both refrigeration and reservoir drain to release liquid which may have entered the chamber bottom. The bottom of the chamber is protected by a base cover. This can be flushed also carefully from the front with water or disinfection solution. Make sure to flush bottom with drain open and prevent overfilling. The maximum reservoir volume is: 3 Liter. What is the temperature control range?

The non-refrigerated version allows temperature control between 5 °C above ambient up to 80 °C. The refrigerated version allows temperature control between 20 °C below ambient up to 80 °C (with a minimum of 4 °C).

What is the temperature uniformity?

The temperature uniformity is ±0.25 °C at 37 °C and 250 rpm.

Are there any limitations in temperature control with specific options installed?

No, also with photosynthetic LED lights running the maximum temperature is 80 °C.

Is there a possibility to increase the chamber humidity?

No, the device is not designed to create a humid chamber environment nor work with a humid chamber environment. The base cover is only for catching potential spillages, protection of the drive and cleaning purposes.

How long does the Balance Master (BM) calibration take?

Balance Master calibration must be performed on one device at a time with no other device running the speed function. The calibration can take up to 60 minutes per device. It is typically much quicker with a single device and with a double stack. Correct installation and leveling is very important for quick and successful Balance Master calibration. Balance Master has to only be done one time during installation or when shaker is relocated to another position.

How long does the Balance Master optimization take?

The Balance Master optimization does take maximum 10 minutes.

What is the benefit of Balance Master?

The Balance Master function assures that the platform is always in perfect counterbalance. Any imbalances will be immediately detected and with Balance Master Optimization the counterbalance can be easily adjusted to optimum. Balance Master facilitates full flexibility in load and speed, reproducible shaking and minimal wear out of shaker components.

How often do I have to do Balance Master Optimization?

Only if the device gives a speed alert. The counterweight is already optimized for the position by the initial calibration procedure (e.g. ground, middle of the stack). It may be necessary to adjust the counterbalance when very high or imbalanced loads are run at high speeds.

Do you recommend to adjust the counterbalance every time after change loads?

No, the shaker is optimized with initial calibration to run most load/speed scenarios. Balance Master Optimization must only be done when the shaker notifies the user of an imbalance (alarm notification via display).

Can I use the Balance Master function when the S44i is stacked with an Innova 44?

Devices only run speed reduced speed: 2 devices stacked: 20 rpm – 300 rpm and 3 devices stacked: 20 rpm – 250 rpm. When using in a mixed combination with Innova 44 we recommend using the tables in the operating manual for initial average adjustment of the counterweight. Calculate the platform weight plus the average amount of weight. Balance Master optimization will not work not optimal as it is not calibrated and may give wrong results

Can I upgrade the Innova S44i with photosynthetic lights?

Yes, the LED light option can be retrofitted. All refrigerated versions can be later upgraded with the LED photosynthetic light option by our Eppendorf Service team.

What is the maximum flask size I can use when the photosynthetic lights are installed?

There is no limitation in flask Size, the maximum flask size stays 5 Liter.

What type of photosynthetic lights do you offer?

We offer LED lights with a maximum light intensity of 150 PAR (µmol m-2 s-1). The uniformity across the platform is ±10%.

What is the minimum and maximum light intensity?

The maximum light intensity is 150 µmol m-2 s-1 or 10,000 Lux. The minimum light intensity is is 15 µmol m-2 s-1 1000 Lux.

Are there any limitations when using the photosynthetic lights?

The unit must be a refrigerated unit to compensate the additional heat in chamber.

What is the average rated life of the LED light and can I replace single bulbs?

The LED light bank was long-term tested for three years at full intensity and in an 80°C chamber and did not show any degradation. Replacement part will be the whole bank.

Is the light intensity adjustable?

The light intensity is adjustable between 10 - 100 % in increments of 1 %. Minimum = 15 PAR (µmol m-2 s-1), maximum = 150 PAR (µmol m-2 s-1). The control and intensity adjustment can be done easily via the main display and requires no extra control element(s).

What spectral range do the lights have?

Are the photosynthetic lights programmable with a timer?

Yes, the integrated programming function offers full flexibility. Different light intensities or on/off phases in a row can be programmed. Cycling of steps or whole programs e.g. to imitate circadian rhythms is also possible.

Can the chamber be shielded from external light?

Yes, the optional window shade can be easily placed in front of the door window to prevent light entering the chamber, e.g. for light sensitive applications or photosynthetic light applications. The window shade can easily be attached and removed again.
