Can the Centrifuge 5427 R be opened in the event of a power failure?

Yes, in the event of a power failure, the unit can be opened by means of the rotor key. Before the centrifuge is opened, always remove the mains plug from the plug socket and wait for the rotor to come to a standstill. For the emergency release of the centrifuge lid, the small plastic cap on the right front corner of the 5427 R has to be removed. Then insert the rotor key into the nut underneath the cap and open the lid latches by turning in anticlockwise direction.

How can the Centrifuge 5427 R be cleaned?

The Centrifuge 5427 R can be cleaned with neutral detergents. For disinfection, we recommend alcohol or alcoholic disinfectants. Make sure that the relevant legal requirements and directives are observed when choosing the disinfection method.

Sometimes my Centrifuge 5427 R displays "EP". What does that mean?"

EP" is displayed when the automatic standby function is active. If the centrifuge remains unused for more than 15 min., the standby function is activated, indicated by "EP" on the display. By using any key, the centrifuge is re-activated and ready to use.

Can the Centrifuge 5427 R automatically recognize an installed rotor?

The Centrifuge 5427 R has an automatic rotor recognition system. A newly installed rotor is recognized at the beginning of the run. The maximum speed of each rotor is automatically limited, if necessary, to prevent over-speeding of the rotors.

What benefits do I gain from the centrifugation of my samples with the swing bucket rotor?

Swing-bucket rotors are particularly advantageous for gradient centrifugation applications as horizontal layers are formed that remain in this position at the end of centrifugation (e.g. blood/Ficoll).

What is the benefit of a so-called Kit rotor?

When working with nucleic acid purification kits with spin columns by certain manufacturers, sometimes tube lids cannot be closed any more. This means you have to centrifuge with open tube lids. These tube lids might rip off during the stress of centrifugation and might damage the centrifuge. In order to prevent this, the kit rotor with the raised rotor rim was designed. Open tube lids, supported against the rim, will not rip off during centrifugation. Another feature of this rotor is the enlarged distance between the rotor bores. The enlarged distance between the bores offers a very convenient handling when loading / unloading the rotor. Especially when working with long spin columns (e. g. from Princeton Separations) you will need a larger distance between the bores to load the rotor fully and make use of the complete rotor capacity.

What tubes can be centrifuged with the Centrifuge 5427 R?

0.5 mL micro test tubes can be centrifuged in all rotors for 1.5/ 2.0 mL micro test tubes using an adapter.

Up to 16x 5 mL micro test tubes can be centrifuged in the aerosol-tight rotor FA-45-12-17.

In which rotor can I centrifuge spin columns?

Spin columns or small filtration columns, for example for centrifugation of DNA or RNA, can be used in all rotors for 1.5/2.0 mL micro test tubes. In particular, however, the kit rotor F-45-24-11-Kit is specially suitable for such spin columns, as open tube lids are supported by the raised edge of this rotor type and are no longer torn off (e.g. during elution of the sample from the column into a micro test tube)

The "Fast Temp" function is a temperature control run of the Centrifuge 5427 R without samples and with a rotor and temperature-specific rotational speed in order to quickly bring the rotor chamber, incl. the rotor, buckets and adapters, to the set nominal temperature. In order that the samples can be centrifuged from the beginning on at the desired temperature, one starts this temperature control run during sample preparation by pressing the "Fast Temp" key. The display shows the current temperature and the g-force (rcf) / rotational speed (rpm) as well as "FT" as symbol for a "Fast Temp" temperature run. You can find a more detailed description in the operating instructions of the Centrifuge 5427 R.

What is the temperature range of the Centrifuge 5427 R?

With the Centrifuge 5427 R it is possible to set the temperature within a range from -11 °C to +40 °C.

Does the Centrifuge 5427 R has an energy saving function?

Yes, with "ECO shut-off" the continuous cooling is switched off if the centrifuge is not used for longer than the preset time. With the default settings, the continuous cooling ends after 8h. In the menu setting "TEMP" the continuous cooling can be limited to 1h, 2h or 4h.

For what applications are aerosol-tight rotors important?

Aerosol-tight rotors are always recommended or even prescribed when working with hazardous or potentially hazardous substances. These can, for example, be viruses, pathogenic bacteria, potentially infectious blood samples or radioactive sample material.

Why are the standard rotors in the micro centrifuges 5418 R, 5420, 5424 R, 5425, 5427 R, 5430 and 5430 R aerosol-tight?

At Eppendorf, the safety of the user always has the highest priority. That is why we now fit our centrifuges with aerosol-tight rotors as standard – at no extra charge.

What kind of cooling liquid is used for the Centrifuge 5427 R?

The cooling system operates with 75g of the "green" cooling liquid R290, also known as propane.

What is the SOFT function?

The SOFT function of the Centrifuge 5427 R allows slower and gentler acceleration and braking compared to the normal acceleration and braking times. This setting is especially suited for applications like e.g. density gradient centrifugation where remixing of phases must be prevented.

Are there any additional safety features implemented due to R290?

Like all our cooling systems, the cooling system of Centrifuge 5427R is a closed system which has no leakage of cooling liquid. For the unlikely case of leakage two additional safety measures are implemeted: All potentially sparks emitting components are shielded, before the centrifuge is ready for operation, a ventilation system takes care to exchange the air in the centrifuge. Therefore, you might notice a few more seconds prolonged initialization phase.

Do I have to take special cautions during transport, storage or installation of the Centrifuge 5427 R due to the green cooling liquid?

No special transport requirements are necessary as only 75 g propane are used. This also means that the centrifuge can be transported by air freight. The room where the centrifuge 5427 R is stored and installed needs to have a volume of minimal 10 m³. In case that more than one device is installed, the requirement stays to 10 m³. During operation leave a safety clearance of 50 cm around the device and at least 30 cm to the rear of the device.

What is the temperature range of the Centrifuge 5427 R?

With the Centrifuge 5427 R it is possible to set the temperature within a range from -11 °C to +40 °C.

How do I recognize aerosol-tight rotors?

Firstly, the aerosol-tight rotors can be recognized by their designation abbreviation “FA” or “AT”; secondly, Eppendorf has color-coded the aerosol-tight rotors. All aerosol-tight rotors have a red rotor lid screw, and a red ring is printed on to the rotor shaft.

Why should I have my centrifuge serviced at regular intervals?

To ensure the optimum performance and precision of your device. This also enables the early detection of any defects which can then be corrected easily. Performing a large number of cycles increases the susceptibility to rotor damage. Any damage can be detected at an early stage during a qualified inspection, i.e. the rotor can be replaced before any serious damage occurs.

Can I carry out software updates myself?

No. Software updates for centrifuges with a USB interface can be carried out easily by a qualified Eppendorf Service technician via the existing USB port. This ensures that safety factors are up-to-date, that the latest rotors are detected and the running of the machine is optimized with the latest parameters.

How often does my centrifuge need to be serviced?

This depends on how intensively it is used and varies depending on the load on the device. However, it is recommended to have the centrifuge serviced at least once a year.

What do you need to look out for before carrying out a service?

The device needs to be decontaminated first. The device must be freely accessible and all samples removed from the device.

How much are the different centrifuge service programs?

The cost depends on the model and on the service that is performed. Please contact your local Eppendorf Service.

What is IQ or OQ and what do I need them for?

Installation and operational qualification; the IQ documents that the device has been delivered and installed correctly. The OQ checks the correct functioning of a device. IQ/OQ ensure that your quality management requirements are met and that your device operates properly and in accordance with the manufacturer’s specifications.

How often does the OQ need to be carried out for a centrifuge?

The company is responsible for technical checks of devices. Eppendorf recommends regular device qualifications depending on the intensity of use, however, they should be carried out at least once a year.

How can I clean my centrifuges myself?

Use a neutral cleaning agent for cleaning. We recommend using alcohol or alcohol-based disinfectants for disinfection. Please ensure that the applicable legal regulations and guidelines are observed when selecting the disinfection method. Is the rotor inspection part of the service programs?

No, it can be booked in addition to service programs and depends on the type of rotor (fixed-angle rotor or swing-bucket rotor).

Shall we close the lid of the refrigerated centrifuge after the centrifugation is finished?

You should close the lid, if using the centrifuge frequently at lower temperatures. When not in use (e.g. over night) switch off the centrifuge and clean the rotor chamber. The lid should stay open to dry.

These natural coolants have a Global Warming Potential similar to CO₂. The refrigerated microcentrifuge Centrifuge 5427 R was designed with high-throughput research applications in mind. With its aerosol-tight 48-place rotor it handles even the toughest challenges in a safe and reliable manner to provide you the efficiency boost you need. With speeds of up to 25,001 x g, this microcentrifuge is fast enough for all your molecular biology applications. All aerosol-tight rotors are equipped with the Eppendorf QuickLock lids for easy rotor lid opening and closing to enable safe centrifugation of hazardous samples. Choose the Centrifuge 5427 R if a 24-place microcentrifuge doesn't meet your needs regarding tube capacity and if you don't need the extra versatility our cross-over Centrifuge 5430 R can offer you.

Natural refrigerant inside!

Centrifuge 5427 R is the first Eppendorf centrifuge with hydrocarbon cooling. Hydrocarbons (HC) are also known as green or natural coolants. The two most commonly used representatives are propane and ethane (known as R290 and R170, respectively). The GWP of HC cooling liquids are more directly equivalent to CO₂. R290 is a commonly used cooling liquid in large commercial cooling systems. Due to their superior efficiency and performance, the amount of green cooling liquids used is significantly lower than that of a similar system with classic cooling by HFCs. Due to these advantages, R290 and R170 are both on the SNAP (Significant New Alternatives Policy Program for substitution of ozone-depleting substances by EPA) list for approved coolants for usage in refrigerated
laboratory instruments such as centrifuges or even ULT freezers.

Disposal of packaging
The packaging of the centrifuge contains different materials, this can include a wooden pallet, cardboard, a plastic dust cover, and some foam parts. Recycling of materials becomes more and more important every day: Are you aware that nearly all cardboard material is recycled in Europe? Our centrifuge packaging cardboard material consists of approx. 50 % recycling material. Please support the global sustainability initiative of recycling valuable raw material by also collecting the freezer cardboard packaging material and disposing of it in the appropriate collection container at your organization. In respect to the plastic dust cover made of LD-PE and the foam parts, we recommend to select a dedicated recycling partner where PE material can be recycled. We suggest you contact your local waste hauler or facility management team to understand the available recycling options for your organization.

Disposal of instrument
Our centrifuges last for many years, but if they need to be replaced, we kindly ask you to fulfill local requirements for disposal of these instruments. We strongly recommend a certified local recycling partner with experience in instruments with active cooling. Keeping it "local" reduces the impact of transportation, and the "certified" aspect is recommended due to the safe and sustainable removal and recycling of the cooling liquids if it is a refrigerated centrifuge.

Decontamination
This piece of equipment was used in a laboratory and/or was used to handle biological samples. Please keep in mind to adequately decontaminate the equipment which needs to be disposed. Check local requirements. For more information, get in contact with your local biosafety officer and/or waste officer. Check if your local recycling partner has special instructions and/or documentation requirements. You may also use the Eppendorf decontamination form sheet as guidance.
